I have a protocol for reverse transfecting 3T3-L1 adipocytes, which differentiates them in DMEM without pyruvate, 25 mM glucose, 10% fetal calf serum (FCS), and 1% penicillin and

streptomycin supplemented with 0.25 mM 3-Isobutyl-1-methylxanthine (IBMX), 0.25 μM dexamethasone, 5 μg/mL insulin, and 10 μM rosiglitazone.

Unfortunately, our media supplier does not have pre-made DMEM without pyruvate. Do I have to make it up from scratch from powder?
