I am working with chimeric antigen receptor and I know the difference in binding without MHC restriction for CARs and know the process of T cell activation and signaling process.
But the thing I am missing is how CARs having for example CD28 or 4-1BB co-stimulation will work compared to TCRs and signaling after CD28 activation and later co-stimulatory molecules .. Are CARs work together with TCRs ?

Not an expert on CARs here but CAR's signaling domain comes from TCR's zeta domain. Costimulatory molecules CD27, CD28, OX40/CD134, and CD137/4-1BB helps drive signaling influencing the proliferation and the persistence of the T cell respectively.
