What is the difference between performing ATPase activity assay and pNPPase activity assay? Why we need to perform it simultaneously?

Why pNPPase activity assay is performed? Only ATPase activity assay is not sufficient?

You are measuring different enzymes:

ATP + H2O -> ADP + Pi
pNP-O-Pi + H2O -> pNP-OH + Pi

Usually, such assays are performed in the context of organelle purification, where these enzymes are then localised in different compartments. pNPPase is a lysosomal enzyme, ATPases are found in plasma membrane and mitochondria (depending on which ATPase we are talking about).
