What will the toxicity levels between Ab42 peptides and fibrils be? Is fibrilisation of ab peptides necessary? Some say peptides are sufficient enough to induce toxicity. Why would that be the case?
A couple of factors to consider. First, preparations of amyloid beta are usually not monodisperse unless a lot of special precautions are taken (for monomers size exclusion chromatography or multiple rounds of seeding for fibers). Second, the distribution of species changes upon incubation. Most toxicity tests are taken after 24 or 72 hours, which is sufficient to form oligomers from the monomeric state. Finally, not all toxicity tests give the same result for a variety of reasons which i can discuss if you are interested.
If you have a relatively monodisperse sample and toxicity assay with a fast read out, generally the monomeric and fiber forms are not toxic but intermdiate size oligomers are.
