One obvious thing to look at is the number of articles published historically- discipline wise and overall. However, it may not give a complete picture unless the quality of publications are factored in somehow.
Is any one aware of published work that has investigated these issues or general growth of scientific knowledge/literature historically?

Knowledge is growing in all disciplines, and usually when a researcher examines this growth he chooses first a discipline or two and observes the knowledge growth in theses fields. Knowledge is not only about papers. There are books (various types), grey literature (technical reports, patents), conference proceedings and more, so, an observation of knowledge growth in a discipline should include all these items.
