I want to check z potential versus pH. What is the best way to check and prepare sample for z potential. For z potential is it necessary to centrifuge the sample or it may analyze before settle down of particles?

Typically you can allow a settling material to settle for a few minutes and take the sample from the resulting supernatant. Typically the zeta potential is broadly independent of size if the chemistry remains the same.
