The following method is used routinely by my students to calculate total soluble carbohydrates.It is the method of Dobios et el ,1956 .the method is as follows:1.take 500mg of ground tissues.2.put into test tubes 90ml capacity and 70 ml of hot distilled water to each tube. and put in water bath for an hour to extract carbohydrates, the n cool the samples to room temperature.3.filter the extract and take the filtrate.4.take 5ml from the filtrate and to it 25 ml of distilled water.5.take 1 ml and add 1ml phenol and 5ml of concentrated sulphuric acid . to cool at room temperature.6. measure the absorbance of the samples at 490 nm. 7.determine the concentration of total soluble carbohydrates by constructing standard curve in which glucose is used. prepare 100mg of glucose in liter, from which prepare five different concentrations.take 1ml from each concentration, and do the same additions as above, and measure the absorbance at 490 nm.Do not forget the dilution factor.
