That really depends on the active compound in a particular plant extract. If a plant produces a non-polar compound, the benzene or acetone may be the best. If the active compound produced by a plant is polar, ethanol or even water is best.
Some people extract the plant in several solvents going from less polar to more polar. As an example hexane -> ethyl acetate -> methanol -> water. Other people extract in ethanol since it can extract non-polar and polar compounds.
I like to use methanol or ethanol and run a column screen which provides a pathway to purify the active compound(s) .
I think you should follow bioactivity guided extraction and fractionation. Firstly the powder will be subjected successive extraction with different solvents in increasing order. All the extracts will be screened for activity. The active fraction may be subjected to column or HPLC to detect active phytoconstituents.
