So far I've only been able to capture 50% of the antigen using incubation at 4 degrees overnight in PBS pH 7

The best capture technique depends upon the particles you are using and the concentration of target. For example, rare cells, using our 250 nm particles require a ratio of 10000 to 1 non targeted to targeted cells with nano particle to cell dilution of 2 x 10^7 cells / 0.5 mg of nano particles. if you are using conjugated particle then it would be better to use solution technique. then centrifuge and collect it back.. i think this will work..
