Hello, I would like to isolate Gliocladium spp. from soil, to evaluate its use as a biopesticide, but I do not find any good protocol for it, does anyone know any method?
Dear Maria del Carmen Casado Munoz, If you are isolating from soil the prevention of mixed cultures might be an obstacle. Gliocladium roseum can be an aerial endophyte on plants such as strawberry, raspberry and soybean. Using raspberry and isolating on potato dextrose agar should be able to get clean cultures. Once you get your isolates you might be able to select for isolates which have resistance to benzimidazole fungicide and then an amended agar would be helpful in assaying isolates for persistence and spread.
