I'm working with scaffolds for bone tissue engineering. MG63, human osteo sarcoma, Saos-2 and MC3T3-E1 are commonly studied cell lines for bone tissue engineering applications. But these cell lines are sarcoma cell lines. Can we use these cell lines for tissue regeneration studies. If so which is the best among these?
Saos-2 behave similar to osteoblast bone forming cells. In addtion, it is useful also to consider the use of stem cells such as human bone marrow stem cells in order to performe differentiation studies.
I prefer primary cell culture rather than transformed cell lines.
Actrually, I agree with Dr. Ernesto. Beltran-Partida, the primary osteoblast is better than the MG 63 and MC3T3E1，but the requirements for the primary cells are even higher than those two cell lines. Besides, I think the bone marrow mesenchymal stem cell is another good choice. The differentiation ability is better than the MG 63 or other cell lines under porper conditions.

If you are going to differentiated cells towards osteoblst via BMP-2 signaling pathway, C2C12 would the best, its BMP-2 expression is negligible and its suitable for evaluating the small molecules and other osteoinductive biomaterial. If it is not your option and you are going to use osteoblats, its primary is the best. You can easily isolated it from the bone.
