Root angles for WT and mutant lines were images, and have been measured every 10 minutes over the time course of the project. I have tried to use an ANOVA with the Tukey post-hoc test but I'm not sure that this is correct. Most of the research papers I have seen have used a students t-test, but i am comparing more than 2 lines, with a different number of replicates per line, over the time course of 0-14 hours.
There is no such thing as a mean angle, so the correct method for measuring and comparing biological angles is to use the following:
Vector mean direction = arc tan S/C, where S = (Σ sine θ)/n and C = (Σ cos θ)/n
and Mean resultant vector length (or ‘strength’), R = (√ [S2 + C2])/n
where θ = angle and n = no. of angles or no. of roots
As Stefano D'Alessandro said, if you can show that a range of these values is normally distributed, you can compare these values by t test or anova, which is just multiple t tests, but with adjustment for the fact there are multiple comparisons :)
