Is 2 weeks of high fat diet too short before STZ injection (low dose, 5 days)? My mice are weaned at 4 weeks and acclimatized to a normal diet 1 week before subjecting them to a high fat diet. While I have not performed GTT before STZ injection, I did observed an increase in blood glucose by week 3 of high fat diet feeding with my preliminary test group. Thanks to anyone who could help. References have varied protocols and rarely more than 2 of them agree so it is kind of hard for me to make a stable model.

Hi Professor Khaled! Thank you for the recommendation! While this paper did not include 2 weeks of HFD administration prior to STZ, it did lead me to another paper that did. I suppose between 2-4 weeks is recommended to induce impaired insulin signaling in rodents prior to STZ injection.

Yes, it answers what I was asking for. I was gonna assume that the effect began at 2 weeks and was maintained until 4 weeks, but like I said I found another paper that reinforces my speculation. Different studies just has varying experimental designs that made it difficult to decide which model we should base our desired model with. I tried to ask for the full text from the author whose paper you suggested for further reading. Thanks again!
