I am planning to use ascorbic acid as a means to reduce phototoxicity while performing time lapse imaging of mammalian cells expressing fluorescently tagged proteins. Can anybody suggest an appropriate concentration?
The question relates to how it is intended that ascorbate is delivered. If orally, then normal daily intake does not generally permit much higher concentrations than 50 uM in plasma. If applied topically as a skin cream then much higher concentrations will be possible 5-10 mM, although care should be taken to avoid metal ion contamination as this can lead to a so-called "pro-oxidant" effect generated by hydroxyl radical formation as ascorbate can take part in Fenton reactions. This can be avoided by co-addition of quercetin around 20-50 uM, which has metal chelator activity and is also an effective antioxidant.
