for this i have taken 0.050g of sample in 10ml of methanol and then 0.3ml from this and added 2.25ml follin reagent and then 2.25ml of saodium carbonate.and got result as 38 ppm from graph But now i am not getting how to calculate my result.
i have taken .05g of freez dried bark sample of plant mixed with10ml of 80% methanol. then this mixture was shaken at 250 rpm for 2 hours. then after centrifugation 0.3 ml of the supernatant was mixed with 2.25ml Folin ciocaltew reagent after 5 minutes then 2.25ml of sodium carbonate (6%) was added. then after 90 minutes the absorbannce of sample and standards (Gallic acid) was taken. and from calibration curve ( which is prepared in the rang 10 to 100ppm Gallic accid) i got the result of my sample as 38 ppm. and i have done triplicate. Now how i will convert my answer in mg/g of Gallic acid?
ic, erm, what i would have done is 2 prepare the standard curve in mg not in ppm and write my results in mg/g gae. But this is what u can do, change the standard (ppm) to in mg. this measurement refers to how much of gallic acid you dissolved in the stock solution.
Yeah Erum, just convert your readings in mg scale, for which as Ikhtifar suggested, need to convert the quantities of standard curve into mg scale too. That should solve the problem.
