I have to prepare plant nuclei slides for light, fluorescent and electron microscopy. What are the best protocol/s for it? What stain/s can be used for it? I know for fluorescent microscopy DAPI can be used, what else? And for electron microscopy there must be some other chemicals to be used. Your suggestions will be of a valuable help.
For fluorescent microscopy u can use Hoechst dye. For electron microscopy.

For mitotic nuclei preparation: Select the meristematic tissue of your plant (root tip or leaf tip). The keep for hydrolysis in 1N Hcl at 60 degree Celsius for 2-5 minutes. ( You need to optimize the time for your plant). Then wash with water to remove the traces of Hcl. Now you can stain with various stains. For light microscopy use withe Basic Fuchsin or for fluorescence microscopy you can use plant DNA specific fluorescent dyes. Hope this should work for you.
