I am interested in ordering a modified DNA oligonucleotide such that it's terminal 3'-OH is blocked in a way that it can be unblocked. Or a dNTP with such a modification that can be added to the 3' end of an oligo. The problem is that this sort of modification is either very rare, or I am uninformed and having trouble finding it. Does anyone have any suggestions for such modifications that are commercially available?
An example would be allyl esters or alkoxylamine groups, or anything that is used in NGS sequencing. In illumina for instance there is a 3'-O blocking group that stops synthesis, the color of the dye is read, and the the blocking modification is removed chemically so that the process can repeat.
