I read a lot of themes on this subject and concluded that recommended % of glycerol for freezing of bacteria on -80 Celsius is 20-40%.

As I understood when you use 40% glycerol after keeping at -80 Celsius content is "partially" liquid.

I want to mix overnight culture with sterile glycerol and put sterile glass beads (pearls) so that cells form biofilm on the beads.

Routinely I would take the cryotube, get one glass bead, role it on the surface of the agar and than autoclave it, or just immediately after use put it back into the cryotube.

My main question is which glycerol % should I make in order to stay liquid after long time storage on -80 Celsius?

Does anyone have similar procedure, with glass beads?

What would be disadvantage of the procedure with glass beads and high % glycerol, if any?
