If I'm trying to isolate a vesicle without detergent, could I possibly use urea and/or guanidinium chloride to facilitate detachment of the vesicle into the cytosolic fraction of cells? Or does the answer to this question depend on the composition of the vesicle (which is unknown at the moment)?

The question is somewhat unclear. Which vesicle is to be detached? Might I understand Subcellular vesicles are concerned? Although the molecular structures of the target vesicles are not determined, it is worthy to try it, using either urea or guanidinium. These chemicals are known as protein denaturing. So, if one of them is used at an appropriate concentration (e.g. 4-5 M) and targeted vesicles contain accessible protein anchors, linking them to cellular supports, that undergo denaturation, the vesicles get dissociation from their supports, due to the anchor's denaturation.
