RLU values are changing daily for the same kit and control in the CLIA assay on fully automated analyser. Can any one help in this issue regarding the significance of variatin of RLU values and any trouble shooting to be taken
RLU stands for relative luciferase/luminescent units. This is calculated after normalizing your raw data to that of the negative control, to demonstrate the fold change in reporter activation in response to treatment.
RLU stands for relative light units. RLUs are the output of the optical measurements that CLIA systems use to then convert to assay-specific analyte concentration units or qualitative interpretations for index (cutoff) assays where:
Final Read (RLU) = Activated Read - Background
Therefore, RLU values (be it for sample, control, or calibrator) should be consistent with minimal variation. In fact, many automated analysers rely on precise RLU replicates to indicate a "good" calibrator curve.
Troubleshooting would depend on the system you are using. Try the basics like replacing bulk solutions, preform fluid flushes, and re-do maintenance.
Can you please confirm the type of analyser you are using and if the variation in RLU is for your samples, QC, or calibrators?
