Today 24 April 2014, Bianchi et al. 2014 (Nature 508, 483–487, doi:10.1038/nature13203) reported that oocyte's "Juno" is the reciprocal receptor of sperm's "Izumo1". Their binding is essential for fusion of sperm and oocytes and subsequently for fertilization. I wonder when we use Izumo1-expressing cells or other Izumo homologues with small electrical current intensity for fusion, would it be useful for SCNT? Any suggestion will be appreciated.
1) If Izumo1 only work for oocyte-sperm recognization but has no function for induce activation, it won't help to activation.
2) Sometimes, delayed activation, that means activate the oocyte several hours later after fusion, will help reprogramming, especially in mouse. If Izumo1 do have activation function, maybe it's better to use it later after fusion.
Thanks Dr Zhong for your answer. If Izumo1-expressing cells used as donors, it might facilitate the fusion using a low electrical current intensity/voltage. That is to reduce the oxidative stress caused by electrical activation of the oocyte. Thanks again for your suggestion.
