Hello you can use 2-5% sucrose solution with addition of 1ml of your culture liquid. Incubate it at 30, 40, 50, 60, 70*C, and than analyse for detection of reducing sugars by Benedict's reagent or any available method like Fehling's test or Nelson-Somogyi method.
1. You need to prove the enzyme intra or extracellular.
2. For enzyme activity calculation you need to analyse also protein content in your culture solution by Kjeldahl method or Lowry method for calculation of your enzyme activity per gram of protein.
