Hi Everybody. I have problem with watergate of NMR for metabolomics study of Saliva. It does not work. I was looking for suggestion how to solve this problem! I read a place which was suggested to dissolve the sample in D2O, then freeze drying it, and dissolve it again in the D2O. Finally, run NMR. Can I do that? What about just freeze drying the Saliva and dissolve it in the D2O and run NMR?

The purpose of the the first dissolution and freeze-drying cycle is to exchange as much hydrogen for deuterium as possible. If you skip that step, all of the protons in the sample will exchange and give you a very large HOD peak. At least one prior dissolution and freeze-drying cycle is required; multiple ones would be even better.
