We are inoculating 1 X 108 TMD8 (B cell lymphoma - DLBCL cell line) cells after mixing it with matrigel (1:1) to right flank of SCID mice. We had ensured that cell viability is above 90% and matrigel we had used is of HC (high concentration) grade. Our observation shows that tumor grew upto 100 -150 mm3 range within 15 days and than suddenly started to disappear. This happened twice. We used same lot of SCID mice for development of another xenograft and that works well. This particular problem is encountered only with TMD8 cell line. Any possible explanation will help. Thank you.
SCID mice retain functional NK cells that may limit post-transplantation growth or mediate rejection of some xenografts. Possible solution in these cases can be in the use of NOD/SCID mice (with or without pre-conditioning by sublethal irradiation) or higher-order immunodeficient mice, such as, e.g., SCID Beige or NSG mice.

and Roman Mezencev . We first tried to pre-condition SCID by giving single dose of cyclophosphamide, i.p. 48 hours prior cell injection. That didn't make much difference so we switched to NOD-SCID. Indeed got good results with NOD-SCID mice. Thank you for helping out.
