I need details to study sleep in earthworms belonging to different functional categories. The problems are: (i) alternative medium to replace soil, (ii) colour of light to be used, (iii) flexible system to fit to the earthworm size (length and width), (iv) time duration of study during day and night, (v) automatic recording device etc. Thank you all in advance.

I don't know anything about C. elegans, but I am not aware of any studies involving megadriles. If you wish to observe the worms without detection use a red light as they do not detect the red wavelength.

Is there any technology available that can detect activity in their giant motor neurons without impeding their daily physical activity, attachchable or detached? That ought to be a direct manner in measuring sleep in earthworms

My lab has developed a running wheel sensitive to the movement of earthworms. We have used it for learning studies, but have also monitored the behavior of earthworms for up to 48 hrs. Some info is available via the link.
