What are the possible causes of the secondary screening of SEREX not work?

I work on the identification of cancer-testis antigens in adult ALL using SEREX. The secondary screening is not working for me even try a new serum and AP buffer, could you assist me to work out why is not working?

is there a possibility to lose the phage?
