What are the merits of logP determination by RP-HPLC over shake-flask method?

Hey there!

I used RP-HPLC for determining logP of some synthetic drug molecules. The major reason being rapid and reproducible determination, which was important considering numerous analogues synthesized. What other advantages RP-HPLC offers over shake-flask method?

The reliability of logP is always subjected to the method used. Besides being handy, the HPLC approach depends on the sensitivity of the method, reference standards, the column type and other interactions described in the Hydrophobic Subtraction Model. I don't see any other advantages in it. I still recommend Sirius GLpKa autotitrator for automated pKa, logP, logD, solubility and dissolution measurement. You can also use the calculated value for screening.
