We have generated multiple mutant lines (ATAF family) to see some drought stress response in order to undestand some protein interaction. Could you please advise and suggest me some methods to obtain preliminary data regarding drought stress assay?
Usually as a result of drought stress ABA produced by the plants. You can analyse the response of the Arabidopsis seed germination in a medium containing ABA. You can measure germination percentage or germination rate in the plant seeds. As an altenative you can apply artificial drought by using PEG and then investigate the germination rate or percentage in Arabidopsis mutant plants. These test usually provide as a fast methods for assaying plant responses to drought which can be done in early stage of plant developments.
Another test can be also done in later stage of plant development. When the plants have fully developed leaf you can investigate the response of their leaf to desiccation. This experiment usually done because it can show you how is the functionality of the stomata of plants which can be related to the capacity of plant to conserve water during drought stress. However still i did not start the drought experiment since i am trying to do BiFC assay to confirm my protein interaction. I could see some interaction in protoplast transient assay.
