Hi, I am looking for good and simple positive controls / marker substances to inject into ex vivo normal human skin tissue samples that give very strong and unique color signals in subsequent histologic paraffin sectioning and haematoxylin and eosin (H&E) staining. Thank you for any ideas.
I don't know your aims, but so called tissue marking dyes are used to stain surgical margins and withstand the processing. Another way is tatoo ink.
Hi Gudrun, thank you for your kind reply. My plan is to inject a marker substance into ex vivo normal human skin, than do histologic paraffin sectioning and haematoxylin and eosin (H&E) staining. In the H&E image the injected substance should give a strong and good color signal to distiguish it from the skin tissue. The aim is to study skin injections (depth, expansion, ...) with a new intradermal injection device. I injected 5% and 10% gelatine before, which resulted in purple infiltrates in the skin in H&E images, however a more clear infiltrate color to distinguish the infiltrate better from the skin tissue in H&E would be favorable.
You can stain your gelatine infiltrate with alcian blue or doublestain alcian/Pas.
