Up to now, I have used a polished steel target, from Bruker, for the analysis of the whole bacterial cell. Apparently, there is a different type of target (ground steel), with a special roughness surface.
Someone could explain me the pros and cons of each target, or the specific applications? Is there a better co-crystallisation ? A more homogeneous sample on the position ? Is-it for a special preparation ?

Ground steel has a regular fine structure on its surface.This enables homogeneous co-crystallization preparations (dried droplet approach).In contrast, polished steel has no specific surface structure, it is flat. This one is well suited for thin layer preparations.

I would use ground steel in your case. Bacterial material will hardly give you a sample for thin player preparation..

Following on Christoph's recommendation, you should use rough steel targets for your application. If you use the polished steel targets, you may fail to achieve co-crystallized droplets drying. We have had issues in the past with the stainless steel targets.
We also found out that Eppendorf tips and tubes worked best with stainless steel surfaces. The problem of not being able to get the droplets dry went away once we started to use Eppendorf tips and tubes...
So if you do not have the rough steel targets, you may want to make sure your pipette tubes and tips are of Eppendorf brand.

someone could help me with a groundsteel 384 target plate BC. It is no longer possible to retain the sample inside the well. The sample spreads everywhere. Does anyone know how to fix it?? Using some solvent especially for washing
