The reagents were mixed in a PCR tube with a final concentrations of 80 μM IAPP in buffer (100 mM Tris buffer, 100 mM NaCl, pH 7.3), 1.6mM ammonium persulfate (Bio-Rad, Hercules, CA), and 160 μM Tris (2,2’-bipyridyl)dichlororuthenium(II) hexahydrate (Sigma-Aldrich, St. Louis, MO). Since IAPP contains a disulfide bond that can interact with zinc if reduced, 192 μM iodoacetamide (Sigma-Aldrich, St. Louis, MO) was used to protect any formed cysteines.

The reaction was conducted by simultaneously irradiating all samples with a 23 watt, 100 watt-equivalent, compact fluorescent warm white light bulb (Lights of America, Walnut, CA) at 10 cm from the samples for 40 seconds. A compact fluorescent bulb was chosen to minimize sample heating. The reaction was quenched by adding equal volume of 2X loading buffer containing β-mercaptoethanol (National Diagnostic, Atlanta, GA). Oligomers were analyzed using the SDS-PAGE protocol outlined in Bio-Rad Mini-Protean 3 (Hercules, CA), with a 4% stacking buffer and 16% resolving gel. The bands were visualized using Pierce SilverSNAP Stain Kit II (Thermo Scientific, Rockford, IL). The gels were scanned with CanoScan D660U (Canon, Tokyo, Japan) and band intensity was analyzed using ImageJ (version 1.43).
