Samples were collected via caudal severance from wild sea bass, stored immediately on ice and then in refrigerator. The intention was to centrifuge them immediately but this didn't happen. We intended to measure cortisol levels using a commercially available ELISA kit (Cayman Chemical).

I think you should give it a try. The steroid hormones are pretty stable, particularly if the sample matrix has been kept cold/frozen. We had plasma samples that went through an accidental freeze/thaw cycle and still found robust, repeatable cort values. Cayman does recommend doing an extraction on your plasma before using it on the plate, so even if you have some crud hanging around in your plasma layer after centrifugation, it should be all cleaned up after the extraction.
