Thank you for replying Grzegorz Boczkaj. I am creating calibration curves using FAME standards and hence it will only give me the FAME concentration in derivatized oils. Does the concentration directly indicate the concentration of Fatty Acids in oils? Or do I have to do any further calculations?

The calibration should include all stages of sample preparation, i.e. you need to prepare a series of free fatty acids calibration solutions and perform with them total analytical procedure - including derivatization to FAME. At the end you need to check the linearity and recovery. An internal standard method is preffered.

Can you help me with one more thing. For calibration, I need to use a series of known amount of fatty acid and internal standard in the solvent. Could please give me an example of how to calculate say 2 serial known concentrations with known amount of fatty acid and internal standard in a solvent? It is really confusing me how to prepare serial known concentration by mixing fatty acid and internal standard together in a solvent. Please Help!
