Urease catalyzes the conversion of urea to ammonium ion and CO2.
Subsequently, glutamate dehydrogenase catalyzes the conversion of alpha-oxoglutarate, ammonium, and NADH to glutamate and NAD+.
The conversion of NADH to NAD results in a decrease in absorbance at 340 nm.
BUN is most frequently measured by the urease –UV method in which the urease reaction is coupled to the glutamate dehydrogenase reaction,
generating NAD+.
