I’m working on the synthesis of mPEG-chitosan nanoparticles and I’ve encountered an issue that I hope someone with experience can help clarify.
I used pre-synthesized mPEG-chitosan polymer 1K 25-60% PEGylation, medium MW , which I dissolved in 0.1 M HCl overnight under continuous stirring. The next day, I adjusted the pH to 5, then added TPP solution dropwise while stirring for another 30 minutes.
After centrifugation, I observed a gel-like pellet forming at the bottom of the tube. I attempted to measure the particle size using DLS, but the pellet did not give any measurable signal. On the other hand, the supernatant showed particle size results. I resuspend the Gel with DI water and sonicate but the same .
Could it be that the nanoparticles are trapped inside the gel pellet, and not dispersing properly for DLS or UV analysis? Also No result by using UV. However in literature they put a UV for it
Has anyone faced this kind of aggregation or gelation during chitosan nanoparticle formation?
Any suggestions, troubleshooting tips, or references would be deeply appreciated. Thank you!
