I am using TRIZOL from Invitrogen to isolate my RNA from bone marrow-derived macrophages. I mix Trizol with chloroform, centrifuge, take the upper phase and add isopropanol to it. The problem is, after I spin my samples, instead of getting an RNA pellet I get a "bubble" at the bottom of my tube. The bubble looks like a drop that is separate from the aqeous/isopropanol mix. The size of this droplet is about 20 ul. After washing with EtOH, the pellet looks like gel. Nanodrop quantification is very low, with ratio 260/230 values
