What is the best concentration of glufosinate ammonium to select transgenic soybeans in greenhouse using spray?
I would like know the best herbicide concentration for transgenic soybean (Glycine max) selection (e.g. 35S:bar or pat genes) in greenhouse condition using glufosinate ammonium (Liberty®, BASF) spray. I've been trying to make a death curve with a national soybean cultivar using water + Silwet L-77 + 64 to 320 mg/L of glufosinate ammonium (Liberty), and spraying when the plants had the first expanded trifolium.

Are those plants in the picture wild-type or transgenic?
Have you finished doing the 'death curve' with the wild-type and find out the best concentration for selection?

In the Abstract, it says that "leaves painted with 135 mg/L Basta could efficiently identify the transformants." I don't think that this journal is SCI, but it is no hurt to try. Beware that different soybean varieties can have influence on the optimum selection concentration. See attached figure (Identification of the herbicide resistance of the leaves of transgenic soybean plants using 135 mg/L Basta).
I am wondering: can 'seed' selcetion (seeds on selection media) be done on soybean, like Arabidposis and tobacco?
Attached is another paper. They also used 'painted leaves with BASTA' method to quickly identify transgenic young plants. It will be good if you include a section of 'wild-type' plants as controls if you use this method.

The selection in culture medium of soybean transgenic tissue is approx. 5 mg/L, but seed screening I never tried. Leaf-paiting from this concentration is ok for me, but I have observed many false-negative or false positive using this approach. Spray can be more practical and efficient.

I agree that spray is simpler, but if 'painted method' can give many false-positives, won't the spray method give many false-postives too? I have not seen many publications using BASTA-spraying method for transgenic soybean selection. Have you seen any of such papers?
I don't have any article, but but I'm trying to establish. I know that have research groups that successfully use this spray method.
we use working 100-120 mg/L concecentration for selection of transgenic plants in Arabidopsis thaliana and it works well.
