Hi, i would like to ask how can i confirm what kind of mass of extract to calculate the total phenolic and flavonoid?
For example, based on calibration curve, i got the concentration of gallic acid is 3.251 mg/mL.
So, by using the formula:
T = C x v/m
I can directly calculate the total phenolic content. But i quite confuse with the mass of extract use. My sample is in fresh condition. For the analysis, 1 mL of sample had been used. But for the extraction preparation , 10g of sample had been used.

For the analysis, 1 mL of sample had been used ? I want to assume that you measured a specific mass of extract from the extract you obtained after evaporation of the solvent you used in extracting the 10g of sample, that is the mass you are to use. not 10g of the plant material. I hope this helps.
