Hi, i would like to ask how can i confirm what kind of mass of extract to calculate the total phenolic and flavonoid?

For example, based on calibration curve, i got the concentration of gallic acid is 3.251 mg/mL.

So, by using the formula:

T = C x v/m

I can directly calculate the total phenolic content. But i quite confuse with the mass of extract use. My sample is in fresh condition. For the analysis, 1 mL of sample had been used. But for the extraction preparation , 10g of sample had been used.
