If you separately create topology for your ligand and your lipid but via same name, the second topology has been added to previous topology file or create absolutely different topology file but remark the previous one ?
-If the answer is different topology, how can I make them become one ?

You can't use topol.top for multiple entities. For complex topologies, make use of the #include mechanism to add parameters/moleculetypes using .itp files.
