I have never tried it for that purpose, but alginate sets when you add calcium.Alginate is used to encapsulate cells. Your bacteria would have to accept the calcium, and should not be able to degrade alginate.

Dear Jakup, we simply grow our psychrophillic bacteria in cooled incubator and if they are anaerobic, grow in anaerobic jar. We do not overlay our plates. It is my suggestion.
