In order to investigate the effect of various growth media on urease activity of S.pasteurii, I want to culture the bacteria in 64 different liquid media. Then I should add a given amount of each culture medium to a specific amount of urea solution and measure the urease activity (@ standard condition). As the biological age of bacteria, which is added to the urea solution, influences its urease activity, I should add each bacteria solution to the urea solution at the same age, for example at the time of early stationary phase. How can I find the time of early stationary phase for the bacteria cultivated in the different media? Should I find the growth curve of the bacteria for all 64 cases?
hi, I am growing mycobacteria in different media and yes the growth rate differs from medium to medium. Sometime the lag phase is influenced as well and it would take longer until the culture reaches the exponential phase. If I would do your experiment I would make sure to capture the bacteria in the mid-exponential phase. You could always make two cultures for each medium of which you leave one longer to see which max OD they reach. I take my samples from OD 0.6-0.8, which corresponds to mid-exponential phase. Surely, the best would be to make growth curves for each and see how the growth occurs, and I would guess if you have e.g different nitrogen sources the growth would be similar. Hope I gave you some ideas.
Hi, i think this is because bacteria need certain requeriments to growth and reproduce. in some media, the bacteria can produce more product than others. I had a similar experience in the growth of Bacillus thuringiensis in different culture media.
