I have recently been working with thermal shift assays and suffering a noisy signal from the dye. The dye I’m using is Sypro orange, and it works fine on the old Bio-rad realtime PCR machine, although it’s very slow. However, as I shift to use this new fancy TSA machine from QIAGEN (QIAGEN®, Rotor-Gene® Rotor-Disc™, it’s super fast), I got a very noisy signal from the dye only. The dye itself gives a “nice” peak at about 50C. Perimeters I set: HRM-yellow or HRM-orange. he gain value is 7-5. Does anyone have experience on QIAGEN®, Rotor-Gene® Rotor-Disc™ ?
