Multicompartment PK–PD models are used by TCI systems to calculate the infusion rates required to reach and maintain the target concentration.
A computer or microprocessor is required to perform the calculations and control the infusion pump.
TCI systems target a concentration in the plasma or at the site of drug effect to allow a stable sedation plan.

Dr. J. B. Dyck is a anesthesiologist and researcher that has directed or collaborated on different clinical studies about dexmedetomidine. The pharmacokinetic model that carries his name, designed for the use of dexmedetomidine for target-controlled infusion (TCI).
The Dyck model combines pharmacokinetic data derived from the studies of plasma concentrations after a bolus dose with data acquired during and after a computer-controlled infusion. However, this is a very preliminary model.
Only the Dyck, Dutta, and Talke models used TCI administration (Dutta and Talke used unpublished models) for model development.
