I'm planning experiments and have no idea about the yield of MDSC induced with GM-CSF and IL-6 from PBMC.
Could anyone who have experience please give me a ballpark estimation?
i.e., Seed PBMC at 0.5M/ml, 5ml/well in 6-well plates, add rhGM-CSF@10ng/ml and rhIL-6@10ng/ml. How many CD33+ CD11b+ MDSC will yield on Day 7 of culture?
It depends on ur skills and how familiar u r with the techniques involved. I dont think anyone can or shd give u an exact number. Do the experiment, measure ur yield and then design ur project based on your personal results to avoid future complications.

The yield in my hands is around 0.15M under the conditions above.
