I'm working with adipose tissue derived MSCs. currently I've faced a problem which have never seen before. Some of the cells start shrinking and die locally only in some area of the plate, next day expands to most of the cells and in two days most almost all of the cells are affected. I tested for mycoplasma, negative, no bacterial and fungi, I cleaned everything, incubator, hood, water bath, centrifuge......... refresh all media and cocktails, and now.......
Does anyone have an idea or faced such a phenomenon before?

How well were the MSCs washed before you plate them? Sudden removal of other stromal cells maybe causing stress to MSCs. Residual lipid can also be a problem. Perhpas its good to maintain some other non-MSC popoulation in parallel. If these survive, you can be sure its MSCs and not media. Hope this helps.
Yes, it was not an infection. most probably because of handling, since the fat droplets from differentiated MSCs seem to be sensitive and loose, it could be also by the strength of digestion and pipetting through the washing steps. one should treat them very friendly. surprisingly, the human MSCs are very resistant in this regard and you can play with them easily. thanks for your comment and attention
