The freeze-thaw method in preparation of cell lysate of human cultured cells for ELISA ?

My cells pellet is preserved in PBS at -80 C. And I am about to prepare cell lysate for ELISA. I want to know how to make sure that the cells are lysed after freeze-thaw cycles.
