I use the BM medium, which was usually for amoeba growth, to cultivate the microorganism in water. The picture is observed by microscope after gram-staining. Can someone please tell why the image is such weird?
@Yanjing Li , you are asking for help with 'Can someone please tell why the image is such weird?' , and I am guessing: With "weird image" you think of the uneven illumination of the field of view upper right limit of the illuminated sphere... 1a) Usually - in cases like this one, more detailed info on how the image was taken/generated, are frankly given by the / a requester without being asked. If you don't mind to tell us,
how the image was taken,
type of microscope (ok: Yes, I know it was a brightfield microscope),
condensor setting, etc., etc.,
then I gladly would be able to point you in the right direction from a 'microscopical' point of view ... 1b) Details of techniques, methods applied, especially with regard to the use of "BM-Medium" in context with your further explicit question, naturally would be really helpful. Controversially, if you mean/meant the 'spatial distribution' of the stained particles or even the staining result as such, the first paragraph also matters.

If, as Wolfgang interpreted, by 'weird image' you're talking about the uneven illumination, to me it seems like you didn't hold the (smartphone?) camera correctly in front of the eyepiece. It might be a good idea to use a dedicated microscope camera (or a different microscope fitted with one).
Regarding the 'uneven illumination' I second 110%...for me it too was clear: smartphone image via eyepiece....(camera target / lens axis center somehow displaced from the optical axis of the eyepiece.)
But perhaps this is of no concern to the requester.... since he didn't reply 'anything' for now...
