I have cell lines where they like to stick together (not adhering to the plate). Anyway to have the cells in culture not stuck together in order to perform flow? Cells are in RPMI or IMDM media. Thanks!
Generally, lab to lab differs slightly but mostly we use BSA 1-2% in staining buffer that helps cells not to adhere each other. Or you can try passing your cells through 70 or 45uM cell strainer wash with BSA containing buffer.
Suspend the cells first in SCSR transport medium, dissociate them by passing through an 18 gauge needle, spin and suspend in growth medium and plate. The medium is described in www.noninvasivetech.com
