squishing or compression of resin (DEAE Anion exchange A 50) while eluting it with 1M NaCl? Suggestions
Why would you? All you'd be doing is potentially rupturing the beads. Don't do it.
What flow rate are you using? Does the same happen when running water?
The resin decreases in volume as the salt concentration increases because the salt shields the electrostatic repulsion between the positively charged resin particles. When the salt is removed, the resin returns to its previous volume.
It's a common occurrence with DEAE-SEPHADEX A50 which is due to the ionic effect of the high NaCl concentration on the charged resin. The resin will swell back to normal as soon as the salt concentration is reduced or removed
Hi Arvind, if the compression of the DEAE in high salt is not affecting your purification procedure or efficiency then you should just accept it.
If it is affecting your efficiency, then you should try more robust DEAE resins as recommended above.
