I'm working on a protocol for determination of total flavonoids in microplate: sample + NaNO2 solution + AlCl3 solution + NaOH solution but at the end precipitates the resulting mixture.
Has anyone ever confronted this experience? How has it been solved? Thank you

It's happens. Try to conduct the assay mixture in appandorf tube then shake it well and left it to be clear. Then take the value of assay from the clear solutions. However the terpidity can affect your results.

I want ask question, Why use of sodium nitrite and sodium hydroxide in total flavanoid content by Alcl3 colorimitric method??
The precipitation usually will go away after sometime. If it is still bothering you try AlCl3 methanol method without nitrazation (Dont add NaNo2). Finally add water or hcl and take the absorbance at 410 nm.
